摘要: 用荧光光谱法研究了小檗碱与DNA的键合作用。发现小檗碱插入到DNA双螺旋中使小檗碱较弱的荧光带(λem=520 nm)的荧光强度显著提高。在pH=3.00附近,小檗碱-DNA体系的荧光增强到原来发射强度的69倍(I/I0)。在pH=3.00和pH=7.20时,测得小檗碱与DNA的本征键合常数分别为3.00×105和2.04×105,二者的键合比分别为每分子小檗碱11.05和4.78个碱基对。

Abstract: The binding of berberine to calf thymus DNA has been investigated by fluoresence spectroscopy, the intercalation of berberine into calf thymus DNA leads to a dramatic enhancement of the photoluminescence of the drug at 520 nm. The fluorescence sensitization by DNA is depended on the pH in solution, maximum in emission intensity is 69 fold (I/I0) at pH=3.00. The intrinsic binding constants between berberine and DNA are determined as 3.00×105 dm3·mol-1(pH=3.00) and 2.04×105 dm3mol-1(pH=7.20) respectively. The estimated site sizes are of 11.05 and 4.78 base pairs at two pH.
