摘要: 本工作用光谱表征、电泳分离及高效液相色谱(HPLC)等方法考察了双锌配合物Zn2(DTPB)Cl4(DTPB=1,1,4,7,7-五(2′-苯并咪唑-2-基甲基)三氮杂庚烷)与DNA的结合及其导致的DNA水解断裂作用。结果表明Zn2(DTPB)Cl4是DNA水解的有效促进剂,以超螺旋DNA作为水解底物,其消失的反应符合准一级速率定律,初步估算其速率常数为2.4×10-5s-1

Abstract: DNA binding and hydrolysis by the di-zinc complex Zn2(DTPB)Cl4 (DTPB=1,1,4,7,7- penta(2′-benzimid- azol-2-ylmethyl)-triazaheptane) were examined with spectral characterization, HPLC and electrophoresis experi-ments. The DNA hydrolytic experiments show that the di-zinc complex is an efficient promoter for DNA hydrolytic cleavage, as confirmed by HPLC analysis of the cleavage products. The estimation of disappearing rate of super-coiled DNA indicates that the approximate pseudo first-order rate constant is 2.4×10-5s-1. The reaction conditions for Zn2(DTPB)Cl4 to exhibit its optimal DNA hydrolytic activity are in the neutral or slightly acidic buffers with lower ion strength. The DNA fragments mediated by the hydrolytic cleaver can be enzymatically ligated and end-labeled. The restriction analysis reveals that pBR322 DNA is preferentially cut by Zn2(DTPB)Cl4 at the sites near bp 570, 1500, 2700 and 4160. What is interesting about these sites is that they consist mainly of multiplet nucleotides A and C.
