基于硝基咪唑衍生物的Cu(Ⅱ)、Ag(Ⅰ)配合物的合成、晶体结构及生物活性

摘要: 设计、合成了硝基咪唑衍生物NN'-二甲基-5-硝基-2,2'-联咪唑(L)的过渡金属配合物[Cu (L)(Ac)2]21)和[Ag (L)(NO3)]·H2O (2)。X射线单晶衍射分析表明配合物1是一个醋酸根桥联的五配位双核Cu(Ⅱ)浆轮笼型配合物。通过元素分析、紫外光谱、红外光谱、固体荧光光谱、电喷雾质谱、摩尔电导率及热重分析等测试手段对配合物2进行了性质表征,初步确定了其结构可能为二配位的线型单核配合物。紫外可见光谱、荧光光谱及黏度法分析表明配合物1与小牛胸腺DNA (ct-DNA)相互作用强于配体及配合物2。以2倍稀释法研究了配体L及配合物的抗厌氧菌活性,发现配体对伴放线放线杆菌(ATCC 29523)的抑菌活性最强(最低抑菌浓度MIC=0.625~1.25 μg·mL-1),这甚至要强于经典抗厌氧菌药物甲硝唑的抗菌性。

Abstract: The copper(Ⅱ) and silver(I) complexes[Cu(L)(Ac)2]2 (1),[Ag(L)(NO3)]·H2O (2) (L=N,N'-dimethyl-5-nitro-2, 2'-biimidazole) has been synthesized. X-ray crystallographic study was used to determine the crystallographic structure of complex 1, which exhibits a five-coordinated dimeric copper-acetato paddle-wheel geometry. Complex 2 was characterized by elemental analysis, UV-Vis, FT-IR, solid fluorescence spectra, thermogravimetric analysis, ESI-MS and conductivity analysis. The structure of complex 2 probably was a two-coordinated mononuclear structural unit of linear configuration. The binding property of L and complexes toward calf thymus DNA (ct-DNA) was figured out via electronic absorption, fluorescence spectra and viscosity measurements. Complex 1 revealed slightly stronger DNA binding capability than 2 and ligand. The antianaerobe activities of L and its complexes were tested by two-fold agar dilution method. The results showed that L displayed favorable activity of anti-anaerobic bacteria against Actinobacillus actinomycetem (ATCC 29523) selectively with minimum inhibitory concentrations of 0.625~1.25 μg·mL-1, even better than the classical anti-anaerobic drug metronidazole.
